This step is also important for biological agents because the agent may exert the best antitumor effect at the MTD. The MTD, however, may not be the same as the optimal biological dose (OBD). Early clinical trials of biological agents demonstrated that immunologic effects occurred at doses lower than Page 9 the identified MTD.

The potential clinical impact of TNF depends on an increased understanding of its cellular mechanisms and tumor cell resistance (Frei and Spriggs, 1989).
